Enzymic Control of the Expression of the X Determinant ( CD 15 ) in Human Myeloid Cells During Maturation : The Regulatory Role of 6 ’ - Sialyltransferase
نویسنده
چکیده
To establish the basis for the reduced expression of the X determinant on leukemic blasts and the changes in antigenic expression that occur during myeloid maturation, the presence on myeloid cells of X and related structures was examined in conjunction with studies on the activities of the glycosyltransferases involved in their biosynthesis. Expression of X and sialyl-X was weak on blasts in comparison with neutrophils despite the presence of the requisite precursor structures. Much higher levels of 3-fucosyltransferase activi ty were found in blasts than in neutrophils when nonsialylated substrates were used. but, whereas the enzyme in neutrophils reacted equally well with 3’-sialylated and nonsialylated acceptors, the enzyme in blasts showed a marked preference for nonsialylated substrates. 6’-Sialyltransferase activity was strong in blasts but was not detectable in neutrophils, whereas a much lower level of 3’-sialyltrans-

متن کاملHuman Leukocyte Antigen-G Expression on Dendritic Cells Induced by Transforming Growth Factor-β1 and CD4+ T Cells Proliferation
Background: During antigen capture and processing, mature dendritic cells (DC) express large amounts of peptide-MHC complexes and accessory molecules on their surface. DC are antigen-presenting cells that have an important role in tolerance and autoimmunity. The transforming growth factor-beta1 (TGF-β1) cytokine has a regulatory role on the immune and non-immune cells. The aim of this study is ...
متن کاملThe Expression of Signal Regulatory Protein-alpha in Normal and Osteoarthritic Human Articular Cartilage and Its Involvement in Chondrocyte Mechano-transduction Response
Signal regulatory proteins (SIRP) belong to immunoglobulin super family (IgSF) and relate to integrin signaling cascades. It has been shown that SIRPa is expressed in a variety of cells including myeloid cells and neurons. In the present study the expression of this IgSF member in articular chondrocytes was investigated. Methods: Using a panel of anti-SIRPalpha antibodies, immunohistochemistry...
متن کاملThe Effect of Monophosphoryl Lipid A on Maturation of DCs from Patients with Acute Myeloid Leukaemia
Background: Generation of monocyte-derived dendritic cells (MDDC) is induced in the presence of GM-CSF and IL-4, and a maturation stimulus is added to the monocyte culture to obtain mature Dendritic Cells (DCs) suitable for therapy. TNF-α is the most common cytokine used for activating DCs and generating mature MDDC either alone or in combination with other cytokines. Objective: To compare effe...

Abstract Background: Various studies indicated that epithelial cells derived from the amniotic membranes of human term placenta have inhibitory effect on components of the innate or acquired immune system. On the other hand, during pregnancy one of the most inhibitory mechanisms of fetal survival maybe though to close contact between AECs and maternal myeloid derived (DCs) that leads to generat...
